چکیده

GSH EXTRUSION IN APOPTOSIS Damaged cells opting for apoptosis actively extrude glutathione in the reduced form as an apical event in the apoptotic signaling (Ghibelli et al., 1995; Van den Dobbelsteen et al., 1996). Apoptotic GSH efflux is very rapid and, as a consequence, cells undergo redox imbalance and become susceptible to oxidative stress due to loss of ROS scavenging power (D’Alessio et al., 2003); moreover, the sudden redox disequilibrium causes the formation of inter-protein disulfides among reactive exposed cysteines, altering cell functions and signaling in a pro-apoptotic fashion (Circu and Aw, 2012).

Apoptosis: from Signalling Pathways to Therapeutic Tools

Apoptosis or programmed cell death is a gene regulated phenomenon which is important in both physiological and pathological conditions. It is characterized by distinct morphological features including chromatin condensation, cell and nuclear shrinkage, membrane blebbing and oligonucleosomal DNA fragmentation. Although, two major apoptotic pathways including 1) the death receptor (extrinsic) and...

Protection against nitric oxide-induced apoptosis in rat mesangial cells demands mitogen-activated protein kinases and reduced glutathione.

Inflammatory diseases such as proliferative glomerulonephritis are associated with the production of nitric oxide (NO), which can initiate apoptotic/necrotic cell death. We studied the role of the p42/44 mitogen-activated protein kinases (MAPKs) and c-Jun N-terminal kinases1/2 (JNK1/2) in NO-evoked cytotoxicity in rat mesangial cells (MC). The NO donor S-nitrosoglutathione time- and concentrati...

Glutathione depletion causes cytochrome c release even in the absence of cell commitment to apoptosis.

We demonstrate here that the release of mature cytochrome c from mitochondria is a cellular response to the depletion of glutathione, the main intracellular antioxidant, independently from the destiny of the cells, i.e., apoptosis or survival. On the one hand, cytosolic cytochrome c was detected in cells where the inhibition of glutathione synthesis led to glutathione depletion without impairin...

A functional genomics analysis of the B56 isoforms of Drosophila protein phosphatase 2A.

Members of the B56 family of protein phosphatase 2A (PP2A) regulatory subunits play crucial roles in Drosophila cell survival. Distinct functions of two B56 subunits were investigated using a combination of RNA interference, DNA microarrays, and proteomics. RNA interference-mediated knockdown of the B56-1 subunit (PP2A-B') but not the catalytic (mts) or B56-2 subunit (wdb) of PP2A resulted in i...

Involvement of Cytochrome P-450 in n-Butyl Nitrite-Induced Hepatocyte Cytotoxicity

Addition of n-butyl nitrite to isolated rat hepatocytes caused an immediate glutathione depletion followed by an inhibition of mitochondrial respiration, inhi- bition of glycolysis and ATP depletion. At cytotoxic butyl nitrite concentrations, lipid  peroxidation  occurred  before  the  plasma  membrane  was  disrupted. Cytochrome P-450 inhibitors inhibited peroxynitrite formation and prev...
